家蚕羧酸酯酶基因家族分析及其对不同杀虫剂胁迫的响应

中文摘要:为探究家蚕Bombyx mori羧酸酯酶(carboxylesterase,CarE)基因家族对不同杀虫剂胁迫的响应方式,利用本地BLAST方法在家蚕基因组中筛选CarE基因家族成员,通过生物信息学分析、系统发育树构建、染色体定位与蛋白保守结构域分析等方法对家蚕CarE基因进行特征分析,并测定家蚕不同龄期与组织中CarE基因的表达量,以及在1/10半致死浓度的吡虫啉、噻虫胺、毒死蜱、辛硫磷、高效氯氟氰菊酯与氯虫苯甲酰胺6种杀虫剂处理后,家蚕中肠中CarE基因的表达量。结果表明:通过本地BLAST方法在家蚕基因组中筛选到8个CarE基因,均含有COesterase保守结构域,其编码区序列长度介于1 299~1 950 bp之间,外显子数量为2~4个,蛋白分子量介于48.84~71.75 kD之间。这8个CarE基因均在家蚕中肠与脂肪体中高表达,且均积极响应辛硫磷、氯虫苯甲酰胺与高效氯氟氰菊酯的胁迫。表明家蚕CarE基因家族成员对不同类型杀虫剂的响应存在显著差异。

Analysis of silkworm Bombyx mori carboxylesterase gene family and response to different pesticides

Abstract:This study aimed to examine the response of the carboxylesterase (CarE) gene family in silkworm Bombyx mori to stress induced by various insecticides. Using a local BLAST search, members of the CarE gene family were identified within the B. mori genome. The identified genes were characterized based on predicted physicochemical properties, phylogenetic analysis, chromosomal mapping, and conserved domain identification. The expression levels of CarE genes were quantified by reverse transcription quantitative polymerase chain reaction (RT-qPCR) across different ages and tissues of B. mori, as well as in the midgut of B. mori exposed to six insecticides (imidacloprid, thiamethoxam, chlorpyrifos, phoxim, chlorfenapyr, and florpyrauxifen-benzyl) at 1/10 of their respective LC50 concentrations. The findings revealed that eight CarE genes were identified through local BLAST screening, all of which contained conserved COesterase domains. The coding sequence lengths of these genes varied from 1 299 to 1 950 base pairs, with exon counts ranging from two to four, and protein molecular weights between 48.84 and 71.75 kD. RT-qPCR analysis indicated that all eight genes exhibited high expression levels in the midgut and fat body of B. mori, demonstrating strong transcriptional responses to stress induced by phoxim, florpyrauxifen-benzyl and chlorfenapyr. The study demonstrated that members of the CarE gene family in B. mori exhibit significant differences in their responses to different types of insecticides.
