中文摘要:为获得对多种柑橘害虫具有良好杀虫活性的生防真菌资源,于浙江省台州市黄岩区柑橘园内采集的柑橘木虱Diaphorina citri虫尸中分离获得菌株MSC-f1,通过形态学特征和分子生物学鉴定方法对该菌株进行菌种鉴定,并对其生物学特性、侵染过程和室内毒力进行测定。结果显示:经鉴定菌株MSC-f1为爪哇虫草Cordyceps javanica,其最适生长温度为25~30℃,菌丝生长和产孢量最适培养基为沙氏葡萄糖酵母浸膏琼脂培养基。通过体视显微镜、扫描电镜和组织切片观察可见,菌株MSC-f1分生孢子易粘结于柑橘木虱的节间膜、体表褶皱处,6 h后萌发侵染,24 h后可侵入血腔、器官,并以寄主为营养体快速生长增殖,最终导致寄主死亡。菌株MSC-f1孢子浓度为1.0×108个/mL时对绣线菊蚜Aphis citricola成虫、吹绵蚧Icerya purchasi成虫、柑橘木虱成虫和柑橘全爪螨Panonychus citri等柑橘害虫均有较好的致死效果,其中对绣线菊蚜的致死中时间LT50最小,对柑橘全爪螨的LT50最大,分别为3.80、4.32、4.96和6.17 d。表明菌株MSC-f1在柑橘害虫绿色防控中具有较大的开发应用潜力。 |

Abstract:In order to obtain biocontrol fungal resources with good insecticidal activity against various citrus pests, the strain MSC-f1 was isolated from the cadavers of Diaphorina citri collected in the citrus orchards of Huangyan District, Taizhou City, Zhejiang Province. The strain was identified with morphological characteristics and molecular biological identification methods, and its biological characteristics, infection process and indoor toxicity were determined. The strain was identified as Cordyceps javanica. Its optimal growth temperature is 25 to 30 ℃, and the most suitable medium for mycelial growth and conidia production is Sabouraud dextrose agar with yeast extract. Integrated observations through stereomicroscope, scanning electron microscope and tissue sections demonstrated that the conidia of the strain MSC-f1 preferentially adhered to the intersegmental membranes and cuticular folds of D. citri. The conidia germinated and invaded 6 h after inoculation, penetrated the hemocoel and internal organs 24 h after inoculation, and then rapidly developed and proliferated by utilizing the host as a source of nutrients, ultimately resulting in host mortality. Bioassays revealed that MSC-f1 (1.0×108 conidia/mL) exhibited significant lethal efficacy against citrus pests including Aphis citricola (LT50=3.80 d), Icerya purchase (LT50=4.32 d), D. citri (LT50=4.96 d), and Panonychus citri (LT50=6.17 d). This implied that the strain MSC-f1 had the fastest action against A. citricola and slowest against P. citri. These findings highlight that this highly pathogenic strain MSC-f1 of C. javanica has strong potential as a biocontrol agent in effectively managing citrus pests. |
