中文摘要:为探究Argonaute (Ago)基因家族在调控二化螟Chilo suppressalis RNA干扰(RNA interfer‐ence,RNAi)效率中的作用,对二化螟的基因组和转录组进行分析,鉴定获得二化螟Ago基因家族成员并对其进行生物信息学分析,并用合成的Ago基因dsRNA饲喂二化螟4龄幼虫,选择二化螟的C‐factor基因作为RNAi的标记基因,分析Ago基因对二化螟RNAi效率的影响。结果显示:在二化螟中鉴定到3个Ago基因家族成员,分别命名为Ago2-1、Ago2-2和Ago5,GenBank登录号分别为OR187405、OR187406和OR187458,开放阅读框序列全长分别为2 742、3 375和2 946 bp。饲喂Ago基因的dsRNA后,二化螟体内go2-1、Ago2-2和Ago5基因均被显著沉默,说明Ago基因的dsRNA可以在二化螟体内有效合成。与单独饲喂dsC-factor相比较,饲喂dsC-factor+dsAgo2-1、dsC-factor+dsAgo2-2和dsC-factor+dsAgo5后,二化螟体内C-factor基因的转录水平均显著增加;且二化螟幼虫的死亡率分别显著降低了23.33、23.33和31.11百分点。表明沉默二化螟Ago2-1、Ago2-2和Ago5基因后显著降低了C-factor基因的RNAi效率,并且显著抑制了C-factor的致死效应。 |

Abstract:This study aimed to investigate the role of the Argonaute (Ago) gene family in regulating the RNA interference (RNAi) efficiency of Asiatic rice borer Chilo suppressalis. Through the analysis of the genome and transcriptome of C. suppressalis, members of the Ago gene family were identified and subjected to bioinformatics analysis. Subsequently, dsRNA targeting Ago genes was synthesized and fed to fourth-instar larvae of C. suppressalis. The C-factor gene was selected as an RNAi marker gene in C. suppressalis, and the influence of Ago genes on RNAi efficiency was investigated. The results demonstrated that three members of the Ago gene family were identified in C. suppressalis, named Ago2-1, Ago2-2, and Ago5, with GenBank accession numbers OR187405, OR187406, and OR187458, respectively, and their open reading frame (ORF) lengths were 2 742, 3 375, and 2 946 bp, respectively. Feeding Ago gene-specific dsRNA resulted in significant silencing of Ago2-1, Ago2-2, and Ago5 in C. suppressalis, indicating effective gene silencing via dsRNA in vivo. Compared with feeding dsC-factor alone, after feeding with dsC-factor+dsAgo2-1, dsC-factor+dsAgo2-2, or dsC-factor+dsAgo5, the transcript levels of the C-factor gene increased significantly. Additionally, larval mortality rates decreased significantly by 23.33, 23.33, and 31.11 percentage points, respectively. These results indicate that silencing of Ago2-1, Ago2-2, and Ago5 significantly reduces the RNAi efficiency of the C-factor gene and significantly inhibits the lethal effect of the C-factor in the C. suppressalis. |
