中文摘要:为明确世界性蛀果害虫梨小食心虫Grapholita molesta谷氧还蛋白3 (glutaredoxin 3,Grx3)编码基因在雄成虫生殖中的潜在功能,采用实时荧光定量PCR技术分析其在不同日龄(初羽化、1、2、3及4日龄)和不同交配次数(交配1、2、3及4次)的雄成虫附腺中的表达特征,并利用RNA干扰技术结合生理生化指标解析其对雄成虫生殖的影响。结果表明:克隆获得了梨小食心虫的Grx3,命名为GmGrx3,该基因编码蛋白具有谷氧还蛋白的典型结构域、催化基序及激活位点。该基因在2日龄雄成虫附腺中的相对表达量显著高于其他日龄雄成虫;经历不同交配次数的雄成虫附腺中GmGrx3的相对表达量均无显著差异。注射dsGmGrx3的3日龄雄成虫与未处理的3日龄雌成虫配对后,雌成虫产卵量及卵孵化率均显著降低,且精包中过氧化氢及丙二醛的含量均显著增加,有核精子活率显著降低,说明储存在精包内的有核精子已严重损伤。表明GmGrx3在梨小食心虫精包中能保护有核精子以对抗氧化应激。

Abstract:To investigate the potential role of glutaredoxin 3 (Grx3) in the male reproduction of oriental fruit moth Grapholita molesta, a major global fruit-boring pest, the expression of the GmGrx3 gene was analyzed using real-time quantitative PCR in male accessory glands across different adult ages (newly emerged, 1, 2, 3, and 4 days old) and different mating frequencies (1-4 matings). RNA interference (RNAi) and physiological/biochemical assays were further employed to assess the impact of GmGrx3 knockdown on male reproductive performance. The GmGrx3 gene was successfully cloned and characterized, encoding a protein with typical glutaredoxin structural domain, catalytic motifs, and active sites. Its expression peaked in 2-day-old male accessory glands, significantly higher than at other ages, but showed no significant variation across mating frequencies. Following RNAi-mediated knockdown of GmGrx3, males exhibited a significant reduction in female oviposition and egg hatching rate after mating. Additionally, levels of hydrogen peroxide and malondialdehyde in spermatophores were significantly elevated, while eupyrene sperm viability was markedly reduced, indicating substantial oxidative damage. These data demonstrate that GmGrx3 plays a crucial role in protecting eupyrene sperm from oxidative stress within the spermatophore, thereby contributing to male fertility in G. molesta.
