水稻二氢吡啶二羧酸还原酶OsDHDPR的抗虫功能分析

中文摘要:为明确水稻二氢吡啶二羧酸还原酶(Oryza sativa dihydrodipicolinate reductase,OsDHDPR)在诱导抗虫性中的作用,利用实时荧光定量PCR技术测定OsDHDPR基因在水稻不同组织以及机械损伤、激素和虫害处理前后的表达情况,分析取食OsDHDPR过表达株系和沉默株系后对二化螟Chilo suppressalis虫重和褐飞虱Nilaparvata lugens卵孵化率的影响。结果显示:OsDHDPR基因在水稻不同组织中均有表达,其中在叶和外叶鞘中的相对表达量较高,在内叶鞘和根中的相对表达量较低。机械损伤以及褐飞虱为害后期均能显著诱导水稻叶鞘中OsDHDPR基因的表达;茉莉酸处理0.5 h和48 h显著诱导水稻叶鞘中OsDHDPR基因的表达,而在处理6 h则显著抑制其表达;水杨酸处理不影响水稻叶鞘中OsDHDPR基因的表达;二化螟为害后0.5~12 h显著抑制水稻叶鞘中OsDHDPR基因的表达。与野生型水稻相比,OsDHDPR过表达株系中OsDHDPR基因表达量显著升高,而OsDHDPR沉默株系中OsDHDPR基因的表达被显著抑制;过表达OsDHDPR显著抑制了二化螟虫重,但不影响褐飞虱的卵孵化率,与之相反,沉默OsDHDPR不影响二化螟虫重但显著降低了褐飞虱卵孵化率。表明OsDHDPR参与调控了水稻对二化螟和褐飞虱的抗性。

Functional analysis of the Oryza sativa dihydrodipicolinate reductase in rice resistance against insect pests

Abstract:To investigate the role of Oryza sativa dihydrodipicolinate reductase (OsDHDPR) in induced resistance against insect pests, the relative expression patterns of OsDHDPR in different rice tissues and under mechanical wounding, hormones treatments and insect infestation were analyzed using real-time quantitative PCR (RT-qPCR). The effects of OsDHDPR overexpressing and silencing on the larval weight of the striped stem borer Chilo suppressalis and the egg hatching rate of the brown planthopper Nilaparvata lugens were also investigated. The results demonstrated that OsDHDPR was expressed in all rice tissues, with relatively higher expression levels in leaves and outer leaf sheaths, and lower levels in inner leaf sheaths and roots. Mechanical wounding and infestation by N. lugens at later stages significantly induced OsDHDPR expression in rice leaf sheaths. Jasmonic acid treatment significantly induced OsDHDPR expression at 0.5 and 48 hours but significantly suppressed it at 6 hours, whereas salicylic acid treatment had no significant effect. In contrast, C. suppressalis infestation significantly suppressed OsDHDPR expression within 0.5-12 h in rice leaf sheaths. Compared to wild-type rice, OsDHDPR expression was significantly upregulated in OsDHDPR-overexpression lines and significantly downregulated in OsDHDPR-silenced lines. Overexpression of OsDHDPR significantly reduced the larval weight of C. suppressalis but had no effect on the egg hatching rate of N. lugens. In contrast, silencing OsDHDPR had no effect on C. suppressalis larval weight but significantly reduced the egg hatching rate of N. lugens. These findings highlight the involvement of OsDHDPR in regulating rice resistance to C. suppressalis and N. lugens.
