北京市房山区葡萄白腐病病原菌鉴定及快速产孢培养基优化

中文摘要:为明确引起北京市房山区葡萄白腐病的病原菌种类,于2021年从该地区采集表现典型白腐病症状的葡萄叶片与果实样品进行症状观察和菌物分离纯化,对分离得到的真菌进行形态学观察和分子生物学鉴定,并进行致病性验证和快速产孢培养基优化。结果显示:共分离得到3株真菌,编号为JGG001、JGG002和JGG003,结合形态学特征以及基于rDNA ITS、Histone、LSUTEF1-α的多基因序列系统发育分析结果,最终确定引起北京市房山区葡萄白腐病的病原菌为葡萄垫壳孢菌Coniella vitis。相较于普通马铃薯葡萄糖琼脂(potato dextrose agar,PDA)培养基,添加10%葡萄汁的PDA培养基显著促进了葡萄垫壳孢菌的产孢能力,产生的分生孢子器数量是普通PDA培养基培养的5~6倍,表明葡萄汁中含有的某些物质能提升葡萄垫壳孢菌的产孢能力。

Identification of the pathogen causing grape white rot in Fangshan District, Beijing, and preparation of a rapid sporulation medium

Abstract:In order to clarify the species of pathogenic fungi causing grapevine white rot disease in Fangshan District of Beijing, samples of grapevine leaves and fruits exhibiting white rot symptoms were collected from the area in 2021 for symptom observation and mycoparasite isolation and purification, and the isolated fungi were subjected to morphological observation and molecular biological identification, as well as verification of pathogenicity and optimization of rapid spore-producing medium. The results showed that three strains of fungi (JGG001, JGG002, and JGG003) were isolated, and their taxonomic status was determined via the phylogenetic analyses based on the rDNA ITS, Histone, LSU and TEF1-α multigene sequences combined with the morphological characteristics. The causal agent of the white rot disease of grapes in Fangshan District was identified to be the grapevine cushion chitin spore fungus, Coniella vitis. Compared with the normal potato dextrose agar (PDA) medium, PDA medium supplemented with 10% grape juice significantly promoted the sporulation ability of C. vitis, producing five to six times the number of conidiophores, suggesting that certain substances contained in grape juice could enhance the sporulation ability of C. vitis.
